    本實驗探究了在不同丁香酚濃度下(0、0.012 5、0.025 0、0.037 5和0.050 0 μL/mL)的模擬運輸實驗對高體鰤(Seriola dumerili)幼魚造成的應激反應和免疫指標的變化。結果顯示:經過8 h運輸后,各組高體鰤幼魚成活率均為 100%;實驗組幼魚血清中魚促腎上腺皮質激素釋放激素(CRH)、魚促腎上腺皮質激素(ATCH)、魚皮質醇(COR)濃度均高于D25和D50組(P < 0.05);高體鰤幼魚肝、鰓中血紅素加氧酶(HO-1)、糖皮質激素(GR)、熱休克蛋白(HSP70、HSP90)基因的表達量呈顯著差異(P <0.05),結合珠蛋白(HP)基因表達量的降低與其他指標一起維持高體鰤幼魚體內的動態平衡。高體鰤體內溶菌酶(Lyz)、鐵調素(Hep)、核因子κB1(NF-Kb1)、腫瘤壞死因子(TNF-α)、白細胞介素-1β(IL-1β)基因的表達量隨著丁香酚濃度的增加總體呈先升高后降低的變化趨勢,補體3(C3)、轉化生長因子β1(TGF-β1)、白介素10(IL-10)基因表達水平的升高反映了運輸時脅迫應激導致機體產生了大量的有害物質,組織相容性復合物1(MHC-1)、干擾素-γ(IFN-γ)基因表達水平隨著丁香酚濃度的增加呈先降低后升高的變化趨勢。綜上所述,丁香酚在高體鰤幼魚運輸中應激和免疫方面的作用主要表現為抑制氧化應激和炎癥從而引起免疫相關基因的變化,并且在D50組內高體鰤幼魚應激反應最小,免疫狀態最佳。因此,丁香酚適宜作為運輸高體鰤幼魚的麻醉劑,在密封袋密閉充氧運輸模式下丁香酚濃度應以0.025 0 μL/mL為宜。[中國漁業質量與標準,2023,13(3):18-25]

    Abstract:

    The stress and immune parameters in juvenile Seriola dumerili at different concentrations of eugenol (0, 0.012 5, 0.025 0, 0.037 5 and 0.050 0 μL/mL) were investigated in the simulated transportation experiment . The results showed that the survival rate of the juvenile S. dumerili was 100% in all groups after 8h transportation. The concentrations of CRH, ATCH and COR in the serum of juvenile S. dumerili were higher than those in the D25 and D50 group (P<0.05). The expression of heme oxygenase (HO-1), glucocorticoid (GR) and heat shock protein (HSP70, HSP90) genes in the liver and gills of juvenile S. dumerili were significantly different (P<0.05), while the expression level of haptoglobin (HP) decreased together with other indicators to maintain the dynamic balance in juvenile amberjack. The expression of lysozyme (Lyz), hepcidin (Hep), nuclear factor κB1 (NF-Kb1), tumor necrosis factor (TNF-α) and interleukin-1β (IL-1β) genes in S. dumerili generally increased first and then decreased with the increase of eugenol concentration. The increased expression levels of complement 3 (C3), transforming growth factor β1 (TGF-β1), interleukin 10 (IL-10) reflected that the body produced a large number of harmful substances due to stress during transportation. The expression levels of major histocompatibility complex 1 (MHC-1) and interferon-γ (IFN-γ) first decreased and then increased with the increase of eugenol concentration. In conclusion, the effects of eugenol on stress and immunity in the transport of juvenile S. dumerili were mainly manifested by the inhibition of oxidative stress and inflammation, thus causing changes in immune-related genes. The stress response of juvenile S. dumerili was the least and the immune status was the best in the D50 group. Therefore, eugenol is suitable as an anesthetic for transporting juvenile S. dumerili, and the concentration of eugenol should be 0.025 0 μL/mL in the sealed bag airtight oxygenated transport mode.[Chinese Fishery Quality and Standards, 2023, 13(3):18-25]
